Conformational States of Vitronectin : Preferential Expression of an Antigenic
نویسنده
چکیده
A difference in recognition of the adhesive glycoprotein vitronectin (also called S-protein. serum spreading factor. and epibolin) by monoclonal antibody 8E6 (Hayman EG. et al. Proc NatI Acad Sci USA 80:4003. 1 983) was investigated using a competitive enzyme-immunosorbent assay and immunoaffinity chromatography. Recognition of vitronectin in serum was approximately 50-fold greater than recognition of vitronectin in plasma. Recognition of vitronectin incubated with heparin. thrombin-antithrombin Ill complex. or heparin and thrombin-antithrombin Ill complex together was 2.5-. 7-. or 32-fold greater. respectively. than recognition of vitronectin alone. Thrombin or antithrombin III by itself did not induce the antigenic change. Factor Xaantithrombin Ill was less effective than thrombin-antithrombin Ill in induction of the change. Dextran sulfate and fucoidan were more potent than heparin in induction of the antigenic change. whereas dermatan sulfate. hyaluronic acid. heparan sulfate. chondroitin sulfate. or keratan sulfate were less effective. Immunoblotting analysis of serum and of vitronectin incubated with thrombin and antithrom-

متن کاملExpression and Antigenic Evaluation of VacA Antigenic Fragment of Helicobacter Pylori
Objective(s): Helicobacter pylori, a human specific gastric pathogen is a causative agent of chronic active gastritis. The vacuolating cytotoxin (VacA) is an effective virulence factor involved in gastric injury. The aim of this study was to construct a recombinant protein containing antigenic region of VacA gene and determine its antigenicity. Materials and Methods: The antigenic region of V...
متن کاملEvidence that conformational changes upon the transition of the native to the modified form of vitronectin are not limited to the heparin binding domain.
Vitronectin (Vn) exists in vivo in at least two different conformational states, the native and the modified form, and these forms have different ligand binding properties. To characterize the molecular events associated with this conformational flexibility, modified Vn was analyzed by competitive ELISA using a panel of conformationally sensitive antibodies with known epitopes. These studies pr...
متن کاملNew insights into heparin binding to vitronectin: studies with monoclonal antibodies.
Vitronectin is a plasma glycoprotein that binds to a variety of ligands. There is considerable debate regarding the dependency of these binding interactions upon the conformational status of vitronectin, the role of multimerization and how the binding of different ligands can change vitronectin's conformational state. We have developed a method of capturing vitronectin directly from fresh plasm...

Mycobacterium avium subsp. paratuberculosis (MAP) is the etiologic agent of Johne's disease in ruminants and there has been a shift in the public health approach to MAP and human diseases like Crohn's disease. The prevention of infection by MAP in ruminants is thought to deter the high impact of economic losses in the level of dairy industry and possible spreading of this pathogen in dairy prod...
